growth | |
treatment | 3 patient groups: healthy (n=10), mild (n=13), severe (n=8) |
extraction | urine samples were desalted and lyophilized, proteins were redissolved in Laemli buffer and concentrated on SDS-PAGE (1 gel band) for in-gel digestion |
separation | |
digestion | Trypsin in-gel digestion of the total urinary protein mixture |
acquisition | MRM acquisition on AB 5500 Q-trap. Monitoring of 24 targeted peptides (corresponding to 7 urinary candidate proteins plus 2 yeast proteins used as internal standards), 25 targeted peptide ions. The corresponding heavy forms (either AQUA or Pepotec synthetic peptides) of the 25 targeted peptide ions were also monitored. Three transitions were monitored for each of the 50 peptide ion forms (total: 150 transitions). |
