separation | Peptides were separated in 45 minutes by a linear gradient of acetonitrile (5 - 35%) and water, containing 0.1% formic acid at a flow rate of 300 nL min-1. |
digestion | Trypsin digestion |
acquisition | For target detection in crude plasma, peptide preparations were analyzed on a TSQ Vantage triple-quadrupole mass spectrometer (Thermo Fisher) in scheduled SRM acquisition mode. SRM acquisition was performed with Q1 and Q3 operated at resolution of 0.7 m/z half maximum peak width, with a retention time window of 240 s and a cycle time of 2.0 s. |
