summary | Targeted measurement of DNA-repair enzymes upon treatment of cells with gamma-radiation. Samples were collected in 5 independent biological replicates per condition. |
contributors | paolo.cifani@immun.lth.se |
publication | unpublished |
growth | Cells were cultivated in RPMI1640 + 10%FBS and L-glutammine, in standard condition. |
treatment | Cells were irradiated using a 137Cs source. 60 minutes after treatment, cells were washed, detached with trypsin, pelletted and frozen. |
extraction | Cell pellets were resuspended in Laemmli buffer, boiled for 5 minutes and loaded on a SDS-PAGE gel. One single slab was excised and proteins were in-gel reduced, alkilated and trypsin digested. |
separation | |
digestion | One single slab was excised and proteins were in-gel reduced, alkilated and trypsin digested. |
acquisition | Peptides were analysed by SRM using a Thermo TSQ Vantage. |
informatics | Skyline was used to both generate the transition list and analyse the raw MS-files. |
