acquisition | Lipid analysis was carried out by ESI-MS/MS after direct infusion with a flow rate of 5 µL/min into a triple quadrupole mass spectrometer (TSQ Vantage, Thermo Scientific). Lipids were detected by selected reaction monitoring (SRM) using the settings adapted from (Welti, R., Wang, X., and Williams, T. D. (2003) Electrospray ionization tandem mass spectrometry scan modes for plant chloroplast lipids. Anal. Biochem. 314, 149-152). MGDG and DGDG were analyzed in positive mode. The ion spray voltage was set to 3500 V and 5 sheath gas was added. SQDG and PG were analyzed in negative mode and the ion spray voltage was set to 3000 V and 20 sheath gas was added. During all acquisitions, the temperature of the ion transferring tube was set to 270 °C and collision-activated dissociation pressure was set to 1.5 mTorr. |
